Telomere-to-telomere assembly of diploid chromosomes with Verkko

alignment-based scaffold assignment to the maternal haplotype and paternal haplotype for the full-coverage Verkko assembly. Almost all chromosomes are a single color, indicating that Verkko scaffolds resolved most chromosomes end-to-end. The only exceptions are in the acrocentrics, where some of the scaffolds could not be assigned due to low mappability and maternal Chromosome 6 and paternal Chromosomes 5 which are each composed of two large scaffolds. Over 99.

The Verkko assembly is consistent with scaffolds all representing a single chromosome bin. Once again, >99.7% of the scaffold bases can be assigned using Strand-seq. Only 2 and 4 Mb of sequence not scaffolded by Verkko could be assigned to the maternal and paternal haplotypes, respectively. The cluster assignment for the maternal haplotype and the paternal haplotype based on Strand-seq data for the HPRC manually curated assembly.

High-throughput telomere length measurement at nucleotide resolution using the PacBio high fidelity sequencing platform - Nature CommunicationsHigh-throughput telomere length measurement at nucleotide resolution using the PacBio high fidelity sequencing platform - Nature CommunicationsThere exist challenges in quantifying the length of individual telomeres at single nucleotide resolution. Here the authors report a method to capture telomeres and accurately analyse their length in human cell lines and patient peripheral blood leukocyte samples using single-molecule real-time sequencing.

Towards a process-based understanding of rifted continental margins - Nature Reviews Earth & EnvironmentTowards a process-based understanding of rifted continental margins - Nature Reviews Earth & EnvironmentContinental margins record the last stages of rifting before a new ocean basin is formed, with their variety reflecting interactions between tectonic, magmatic, sedimentary and hydrothermal processes. This Review provides a process-based understanding of rifted margin formation, gaining insights into their resource potential for the energy transition.
